If by this you mean 'which villain has only appeared once so far,' I'd go with either C or D.

You got 1,2, and 5 right. On three Zap is made up because Hobgoblin has electric gloves. On four the answer is Beetle, He apeared in more than one comic. On five I meant 'Who did Peter Parker Fight First' That is the Vulture.
